Walgreens.com says that to test for PPD or tuberculosis, a fluid containing the tuberculin organism must be injected under the skin. When a person is injected with the TB bacterium, they get a hard, raised spot on their skin. After 48 to 72 hours, the injection site is checked on the people who took the test. The health care provider checks the injection site and decides if the patient needs more tests for TB, such as chest X-rays and blood tests.
